Library Preparation Services

Overview

Libraries must be prepared before they can be run on the Illumina systems. We accept both prepared and unprepared samples for sequencing.

Prepared Libraries

If your libraries are already prepared, we expect them to run on the MiSeq or HiSeq2500. DNA Libraries should be gel purified and OD'd by spec after purification. We also request a picture of an aliquot run on a 2% TAE agarose gel with 100bp ladder markers. Alternately, you may provide the results file from an Agilent Bioanalyzer, with your peaks and concentrations clearly marked. Please dilute samples to 20nM prior to shipping.

Library Preparation Services

Genomic DNA

We currently offer Genomic DNA Sample preparation using the TUCF protocol below. Please provide at least 5ug (10ug preferable) of high quality genomic DNA (260/280 ratio of 1.8). We will accept a smaller quantity of DNA if a fluorescence-based assay or Bioanalyzer is used to estimate concentration. Please submit an image of your DNA run out on a gel with 1kb markers; there should be no material below 10kb.

RNA

We offer an RNA-Seq sample preparation service using the NuGEN Ovation RNA-Seq System for low input (10-100ng) quantities of RNA, and Illumina TruSeq RNA library preparation for >1ug of RNA. If possible, please validate your libraries with a Nano or Pico RNA chip on an Agilent Bioanalyzer.

Targeted Sequence Capture and Exome Capture

We offer Human Exon capture using either the NimbleGen SeqCap EZ platform or Agilent SureSelect XT platform. In addition, we can help design targeted sequence capture experiments using either of these platforms.
